概要 | This study was designed to investigate the inhibitory effects of 18 commercially available antidepressants on ATP-induced contractions in guinea pig urinary bladder smooth muscle (UBSM) to determine whether they induce voiding impairment. Concentration-response curves for ATP-induced contractions in guinea pig UBSM strips were generated in the absence or presence of selected antidepressants. When inhibitory effects indicated competitive antagonism, pA2 values against ATP were calculated and compared to blood antidepressant concentrations. ATP-induced contractions were competitively antagonized by duloxetine (a serotonin and noradrenaline reuptake inhibitor) and aripiprazole (a dopamine partial agonist). The antagonistic actions of duloxetine and aripiprazole were exerted at concentrations exceeding clinically achievable blood levels based on their pA2 values (5.55 and 5.85, respectively). Other tested antidepressants also did not inhibit ATP-induced contractions of guinea pig UBSM preparations within the concentration ranges used in clinical practice. Antidepressants prescribed in Japan may produce voiding impairment; however, their actions are not mediated via inhibition of UBSM contraction induced by ATP. |
