概要 | Steroidogenic acute regulatory protein related lipid transfer domain containing 10 (STARD10) is a member of the START domain containing lipid transfer protein family. We have previously shown that STARD10 is highly expressed in the liver and involved in bile acids homeostasis. When fed a high fat diet, the liver of Stard10 knockout (Stard10-/-) mice accumulated significantly less cholesterol and triglycerides than wild type (WT) mice. The aim of this study was to clarify the role of STARD10 in lipid accumulation in the liver. The area of lipid droplet (LD) of Stard10-/- mice fed with normal diet was significantly smaller than those of WT mice. We examined the effect of Choline-deficient diet (CDAA), which is known to induce liver specific fat accumulation and nonalcoholic steatohepatitis (NASH) in Stard10-/- mice. The liver of Stard10-/- mice was smaller and the area of LD was significantly smaller than those of WT mice. Gene expression levels of IL-1β and TNF-α were lower in Stard10-/- mice. Recentry it was reported that STARD10 interacts with lysophosphatidylcholine acyltransferase 1 (LPCAT1) to facilitate trafficking of phospholipid from the ER to the lamellar bodies in pulmonary epithelial cells. We confirmed the interaction of STARD10 with LPCAT1 and their localization at the surface of LD in tsA201 cells. These results indicate that STARD10 is involved in transport of phosphatidylcholine in LD formation. |
